Comment:
Plasmodium vivax in thick film - trophozites - note basophilic stipling and Schuffner's dots and ameoboid form of parasite- all characteristics of this species. Because the thick film takes longer to dry than a thin film, the nucleus has time to contract to a bead, in a resting state but the cytoplasm is slower to respond and P. vivax parasites are the most active of the human malarial parasites so the cytoplasm still appears dispersed and often fragmented because the pseudopods have been torn away as the red cell was lysed. Hence it depends on the way the film is made, the thickness and the time it takes to dry, as to the appearance of the parasites. The basophilic stipling represent accumulations of rRNA and are always pathological, caused by ineffective heme formation.
